Time-Resolved Fluorescence (TRF) takes advantage of a particular characteristic of the elements called lanthanides. These fluorophores have long emission lifetimes that allow for changes to be made regarding when detection starts and how long emission is measured.
Time-Resolved Fluorescence Resonance Energy Transfer (TR-FRET) incorporates lanthandies as donors in a unique form of resonance energy transfer (FRET) . Over time the TR-FRET response compares the lanthanide donor signal and the signal from a flurophore acceptor and a ratiometric calculation can be performed. Some common FRET assays are commercially available as the brand names: HTRF®, LanthaScreen®, LANCE®, and Transcreener® ADP2 TR-FRET Red Assay.
Time-resolved fluorescence detection with BMG LABTECH microplate readers
TR-FRET can be performed on the PHERAstar® FSX, CLARIOstar®, and Omega Series readers which are certified for HTRF®, LanthaScreen®, and Transcreener® ADP2 TR-FRET Red Assay. A high-intensity xenon flashlamp provides high performance measurements, while assay-specific filters or optic modules select the specific wavelengths, yield higher light transmission, and minimize unwanted background noise. The PHERAstar FSX offers further TRF-dedicated features such as the TRF-laser and theunique Decay Curve Monitoring function which provides the data needed to use our unique Integration Time Wizard in MARS.
